The objective of this study was to determine the objective response rate in patients with platinum-sensitive and platinum-resistant recurrent ovarian cancer to treatment with trabectedin (Yondelis((R))) administered as a 3-h infusion weekly for 3 weeks of a 4-week cycle. We carried out a multicentre Phase II trial of trabectedin in patients with advanced recurrent ovarian cancer. Trabectedin (0.58 mg m(-2)) was administered via a central line, after premedication with dexamethasone, to 147 patients as a 3-h infusion weekly for 3 weeks followed by 1-week rest. Major eligibility criteria included measurable relapsed advanced ovarian cancer and not more than two prior platinum-containing regimens. Patients were stratified according to the treatment-free interval (TFI) between having either platinum-sensitive (>/=6 months TFI) or platinum-resistant disease (<6 months TFI)/platinum-refractory disease (progression during first line therapy). In the platinum-sensitive cohort, 62 evaluable patients with measurable disease had an overall response rate (ORR) of 29.0% (95% CI: 18.2-41.9%) and median progression-free survival (PFS) was 5.1 months (95% CI: 2.8-6.2). Four patients with measurable disease per Response Evaluation Criteria in Solid Tumours (RECIST) criteria had no follow-up scans at the end of treatment. In the platinum-resistant/refractory cohort, 79 patients were evaluable with an ORR of 6.3% (95% CI: 2.1-14.2%). Median PFS was 2.0 months (95% CI: 1.7-3.5 months). Two patients with measurable disease per RECIST criteria had no follow-up scans at the end of treatment. The most frequent (>/=2% of patients) drug-related treatment-emergent grade 3/4 adverse events were reversible liver alanine transferase elevation (10%), neutropaenia (8%), nausea, vomiting, and fatigue (5% each). La firma argumenta su valoración en el Yondelis, que aunque sólo aporta 1,57 euros por acción a su valoración del grupo, "ha conseguido devolver la atención de los inversores al valor y dotar de una mayor credibilidad al potencial de desarrollo de la cartera de compuestos de la compañía ante la comunidad financiera", la importante cartera de productos en desarrollo y el hecho de que Zeltia pueda ser el objetivo de alguna operación corporativa a medio plazo.

La estructura de fondo de mercado mencionada está en vías de romperse al alza, hipótesis reforzada por una aceleración de compras de inversores institucionales en los últimos meses, por lo que mantendríamos nuestras posiciones largas con un caracter de inversión de largo plazo, pues el objetivo estructural del valor se sitúa muy por encima de los precios actuales.

ABSTRAC :
Saframycin A (SFM-A), produced by Streptomyces lavendulae NRRL 11002, belongs to the tetrahydroisoquinoline family of antibiotics and shares a core structurally similar to that of Ecteinascidin 743 (ET743), which is a highly potent antitumor drug isolated from a marine tunicate. In this study, the biosynthetic gene cluster for SFM-A was cloned and localized to a 62 kb contiguous DNA region. Sequence analysis revealed 30 genes that constitute the SFM-A gene cluster, encoding an unusual non-ribosomal peptide synthetase (NRPS) system and tailoring enzymes as well as regulatory and resistance proteins. Substrate prediction and in vitro characterization of the adenlyation specificities of this NRPS system supported that the last module acts in an iterative manner to form a tetrapeptidyl intermediate and the co-linearity rule does not apply. Although this is different to those proposed for the SFM-A analogs SFM-Mx1 and safracin B (SAC-B), based on the high similarity of these systems, it is likely they share a common mechanism of biosynthesis as we describe here. Construction of the biosynthetic pathway of SFM-Y3, an aminated SFM-A, was achieved in the SAC-B producer (Pseudomonas fluorescens). These findings not only shed new insight into tetrahydroisoquinoline biosynthesis, but also demonstrate the feasibility of engineering microorganisms to generate structurally more complex and biologically more active analogs by combinatorial biosynthesis.

"La salida de este rango otorga una proyección teórica al alza o a la baja de 1,2 euros lo cual hace el valor muy apetecible caso de romper por su parte alta", señalan. Su recomendación es la de 'comprar' con cierres por encima de 8,15 euros.
En estos momentos, no obstante, las acciones de Zeltia retroceden un 0,89% y cotizan en los 7,8 euros.
